The CD163 gene encodes a protein known as cluster of differentiation 163 (CD163), which has a molecular weight of 130 kDa. It is the hemoglobin-haptoglobin complex's high-affinity scavenger receptor. The CD163 protein is made up of a single transmembrane segment, a cytoplasmic tail with multiple splice variants, and an extracellular domain with 1048 amino acids. Innate immune sensing for both gram-positive and gram-negative bacteria is carried out by CD163. During inflammation, activated monocytes and macrophages release the extracellular domain of CD163, also known as soluble CD163. This domain is found in plasma and cerebrospinal fluid. Rapid phagocytosis of the Hb-Hp complex by CD163-expressing macrophages leads to hemoglobin recycling pathway degradation via heme oxygenase-1 (HO-1) production of anti-inflammatory metabolites. Thus, the anti-inflammatory response is indirectly triggered by CD163.
