Exosomes are composed of a variety of membrane proteins, including tetraspanins (CD63, CD81, CD82, CD53, and CD37), endosome-associated proteins (Alix, Tsg101, and Rab proteins), and lipid raft-associated proteins (glycosylphosphatidylinositol and flotillin). The proteins contained within exosomes can be modified and engineered to confer new and desirable characteristics upon the exosomes. For example, molecules can be genetically fused to the exosomal transmembrane proteins, expressed on their surface, and endowed with a range of physiological properties, including the ability to target, demonstrate elevated expression levels, provide improved solubility and prolonged half-life, and elicit antigen immunogenicity.
